4 Australian Methylation & Genomics Summit, Sydney 2018 Speakers Dr Stephanie Fryar-Williams Keynote speaker MB BS BSc (Biochem/Pharmacol) FRANZCP, is a Child and Adolescent and Adult Neuropsychiatrist who undertook the Mental Health Biomarker Project stage 2. ( ) as an Honorary Research Fellow at the University of Adelaide and Director of Youth-In-Mind Research Institute, South Australia, via Basil Hetzel Institute for Translational Health Research and the Queen Elizabeth Hospital, Woodville South Australia. Her research investigates ways that will assist clinicians to predict and confirm the diagnosis of schizophrenia and schizoaffective disorder. is the founder of MTHFR Support Australia. MTHFR Support Australia has become one of the top rating MTHFR and methylation sites in the world. In 2016 Carolyn launched her own range of MTHFR and methylation supplements, wrote her online course on MTHFR and preconception and is currently writing her own book. Dr. Pratt-Hyatt Matthew Pratt-Hyatt received his PhD in cellular and molecular biology from the University of Michigan. How these vitamins connect and inversely correlate with noradrenalin and adrenalin levels. An interesting, meaningful relationship exists between B vitamins, folate, vitamin D and MTHFR. PH.D Upstream cellular mechanisms that include redox balance, cellular energetics and inflammation profoundly contribute to depressive states. Typical SSRIs inhibit the reuptake of serotonin but also act to inhibit a key enzyme, nitric oxide synthase (NOS) needed for synthesis of nitric oxide. NOS plays an essential upstream role in the nervous system. Nitric oxide in the presence of excessive superoxide radical leads to formation to a far more toxic radical species, peroxynitrite. In turn, this leads to inactivation of key neurotransmitters, a situation which has huge ramifications for mental health. Learn how to correct upstream processes in the depressed patient. Our bodies contain a biochemical reflex - an autonomic response - to toxins, stress and inflammation that destroys tryptophan levels. This phenomenon is driven by epigenetic signals from our endocrine and immune systems which help our bodies make more NAD at the expense of calming neurotransmitters. It is becoming more common for doctors and patients to look at MTHFR pathways in the treatment of depression. Research has proven that MTHFR is a risk factor for depression, yet we need to understand how the folate molecule impacts our neurotransmitter levels. Dr. Rostenberg will elaborate on the shared pathways between BH4, DHFR, MTHFR, and folate inside of our nervous system. Dr. Pratt-Hyatt The clinical significance of the organic acids test on mental health Improved technology has brought the realization of personalized medicine closer to a reality. The goal of personalized medicine is to have custom designed treatment plans for each l patient. Two of the most important tools to make this new frontier of medicine functional are the Organic Acids Test (which is a metabolic assessment of overall health) and genetic screening. These two tests work in tandem to help practitioners understand what is happening in the patient s body by assessing the results of the Organic Acids Test (OAT), and then using a genetic test to determine the best course of treatment. These two tests are imperative in assessing neurotransmitter metabolism and the methylation pathway. For the methylation pathway we recommend looking at ten different genes. These include MTHFR, MTR, MTRR, CBS, BHMT, AHCY, NOS3, SHMT1, SUOX, and VDR. There are complementary nutritional markers assessed in the Organic Acids Test that help these enzymes work more efficiently, including vitamins B6 and B12. The neurotransmitter biosynthesis and metabolism pathway is supported by the methylation pathway. Three of the best markers in the OAT for measuring neurotransmitter metabolism are homovanillic acid (a dopamine metabolite), vanilymandelic acid (epinephrine/norepinephrine), and 5-HIAA (serotonin, marker). These markers are the metabolites of the neurotransmitters by the enzymes MAOA and COMT, two genes which possess common polymorphism. Deficiencies in these enzymes due to faulty SNPs may cause low neurotransmitter levels, which may also be caused by low amounts of precursors, cofactors, or increased inhibitors. Assessing these two pathways through an Organic Acids Test and genetic screening can be very beneficial for improved health. 5
