Graduate student UAB, Laboratory of Dr. Victor Darley-Usmar Protein Modifications and the Response to Oxidized Lipids in Cardiovascular Cells Determine how the proteome is modified by endogenous lipid peroxidation using tagged lipids. Elucidate the effects of heme induced lipid peroxidation on mitochondrial function, autophagy and cell death. Examine the role of reactive lipid species on coordinating cellular processes such as upregulation of antioxidants and response to stress.
2 Research Assistant January 2005-April 2005 UAB, Department of Microbiology Used homologous recombination techniques using a suicide vector and conditionally replicating plasmid to delete the gene encoding the protein mpos, a hypothetical porin expressed in Mycobacterium Smegmatis, in the strain SMR5 (wild type M. Smegmatis). Designed primers for Southern Blot, PCR, and sequencing Purified chromosomal DNA from M. Smegmatis for PCR and Southern Blot Research Chemist I November 2003-January 2005 Biological & Physical Space Research Lab UAH, Marshall Space Flight Center Designed clones for overexpression of recombinant proteins. Pyruvate Dehydrogenase complex proteins were isolated and various protocols used for crystallization. Constructed and maintained a database with clone information, such as DNA sequences, primer design, protein domains, and hyperlinks to more information. Created protocols for the cell culture of myosarcoma and lung fibroblast cells obtained from ATCC, for use in radiation studies. Developed protocols for the studying the expression profiles of specific proteins. Used mrna obtained from myosarcoma cells and produced labeled cdna using RT-PCR. Performed microarray hybridization and analysis. Visiting Research Assistant June 2003-September 2003 Lab for Structural Biology University of Alabama in Huntsville Used genetic engineering techniques such as PCR, restriction digest, ligation, DNA purification, and transformation to express recombinant proteins. Both E.Coli and yeast expression systems were used. Clones were screened for expression levels of the protein Cathepsin K (human and the mouse homolog). Research Assistant June 2002-June 2003 Natural Products Drug Discovery Lab University of Alabama in Huntsville Took crude plant extract of the legume erythrina lanceolata from the Monte Verde cloud forest of Costa Rica, and performed several extraction and purification trials using organic solvent extractions, column chromatography, HPLC, and centrifugal TLC.
