For this reson sel integrity remins intt even t elevted tempertures during. To illustrte the dvntge of our two-omponent tehnology we hve ompred evportion from stndrd PP pltes nd FrmeStr pltes: Eh well of non-skirted 96well plte (PP) nd Frmestr non skirted design plte (ode 4ti-0710) ws filled with 10μl HO. The pltes were seled with q dhesive sel (ode 4ti-0560) nd the weight of eh plte ws mesured efore nd fter performing (30 yles x C; C). Tle 1 shows tht the verge volume loss from stndrd PP pltes using dhesive seling ws.3μl per well, whih is equl to 3% of the totl retion volume. In ontrst the volume loss from FrmeStr pltes ws only 0.49μl per well, so less thn 5% of the totl retion volume. 3 Evportion from stndrd PP pltes is highest in the outer wells Sine therml expnsion nd movement of wells in stndrd PP pltes is enhned round the edges of the pltes (see Figure 1) evportion is highest from the two outer rows of wells. Figure 3 illustrtes the levels of smple evportion from different res of PP pltes. Only the inner 3 wells of stndrd 96well plte show low levels of evportion, smple loss is high from the two outer rows whih ontin more thn 65% of the wells. FrmeStr -omponent pltes improve onsisteny of results We hve ompred the degree of evportion from different res of stndrd PP nd FrmeStr pltes. First, the 64 outer wells (two outer rows) of oth plte types were filled with 10μl HO, pltes were then seled with q dhesive sel (ode 4ti-0560) nd their totl weight determined efore nd fter. The experiment ws repeted with set of pltes of whih the inner 3 wells (green re in Figure 3) were filled. Tle shows tht evportion from outer wells of stndrd PP pltes ws signifint nd 65% higher thn from inner wells. In ontrst, FrmeStr pltes showed low levels of smple loss ross the plte. The results elow show tht retion volumes remin onsistent ross the 96wells (or 384wells, dt not shown) in FrmeStr pltes. In ontrst, the retion volumes in stndrd pltes differ inresingly etween wells during. Buffer onentrtions in outer rows will inrese drmtilly nd result in redued enzyme tivity. Improved Seling FrmeStr tehnology provides muh-improved level of sel integrity: During, with het sels or dhesive sels, the exessive therml expnsion/ontrtion of stndrd polypropylene pltes, n hve dmging effet on sel qulity. This is espeilly evident round the edge of the plte where these fores re t their mximum resulting in signifint smple evportion from the outer wells. In ontrst, the higher therml stility of FrmeStr pltes results in relile sel in ll res of the plte during nd in old storge down to -80ºC. The results shown were otined using the following protool with 5μl retion volumes, pltes seled with dhesive sel sheets (4titude ode 4ti-00), in Thermo Px Cyler: 1min 94ºC, 30x(30se 94ºC, 30se 55ºC, 30se 7ºC), 5min 7ºC, holding step t 4ºC.
