RNA-seq data analyzed using the Bioconductor statistical suite. Methylome characterization of single bovine in vivo pre-implantation embryos by RRBS and WGBS. Characterization of global loss of imprinting in embryo and fetal development induced by assisted reproduction in domestic animals. 1
2 Effect of sub-lethal Hydrostatic Pressure (HHP) on transcriptional profiles of bovine in vitro-produced (IVP), vitrified blastocysts. Genes that changed expression due to HHP treatments were identified using Microarray and real-time quantitative RT-PCR, Data were analyzed using GeneSpring 12.1, Bioconductor statistical suite and SAS program. Roles of Brm and Baf170, components of chromatin remodeling complex, during reprogramming using gene-targeting approaches. Reprogrammed somatic cells by the ipsc technology using shrna-mediated knockdown in mouse ips and ES cell lines, ipscs characterization and cell culture. Investigated PRNP (gene caused mad cow disease) and Y chromosome gene targeting in bovine somatic cells using transcription activator-like effector and nuclease (TALEN) technique and CRISPR-CAS9 system. Transcriptional profiling of imprinted, X-linked, histone, mitochondrial and housekeeping genes in mammalian pre-implantation development. Data mining from public databases. Co-discovered the role of Jak/Stat3 signaling in epigenetic regulation of somatic cell reprogramming to Naïve pluripotency. Co-discovered the role of Akt in Stat3 activation and the isoform-specific roles of Akt in induced pluripotent stem cells (ipscs) generation. Generating and characterizing pluripotent bovine ipscs. Sequenced the complete mitochondrial genome of Oreochromisco aureus and analyzed their molecular phylogenesis. Studied the function of mouse gonad-specific expression gene. Co-investigated the genetic diversity between sexes of genetic improvement of farmed tilapia by Simple Sequence Repeat analysis. Teaching and Supervising Experiences: Teaching assistant for ANSC 3323/5621 (Animal Embryology and Embryo Biotechnology) in Department of Animal Science, UCONN, every fall since Training fellow graduate and undergraduate students on molecular cloning, ipsc generation and characterization and module analyses of RNA-seq data. Lab management: supply orders, material transfers, lab cell line storage/maintenance, research protocol development, lab instrument log, user training, and annual maintenance.
